Validation of a rapid semi-automated method for monitoring alcohol abuse by phosphatidylethanol (16:0/18:1) LC-MS/MS determination in dried blood spots

Legrand-Brault et al.

The finding, in our words

A semi-automated LC-MS/MS assay for measuring phosphatidylethanol 16:0/18:1 from dried blood spots was validated across a 10 to 1000 ug/L range with acceptable precision and accuracy. Application to 346 clinical patient samples successfully identified 67 individuals exceeding the 200 ug/L threshold for chronic alcohol consumption.

Liquid Chromatographic Methods for Microsampling in Sports Drug Monitoring: Progress in DBS and VAMS

The analysis indicates that dried blood spots and volumetric absorptive microsampling enable minimally invasive monitoring of prohibited substances, with volumetric absorptive microsampling offering improved quantitative reliability over traditional dried spots.

A validated method for capillary phosphatidylethanol 16:0/18:1 quantification with two different 10-µl volumetric absorptive microsample devices in the same setup

This study validated a method for measuring the alcohol biomarker phosphatidylethanol using two volumetric absorptive microsampling devices, finding that the Mitra device met all validation criteria and agreed with venous sampling. The Capitainer device was deemed suitable but showed reduced accuracy at higher concentrations, with a small proportional negative bias compared to venous results.

Patient centric blood sampling and analysis for diagnostics and laboratory medicine

A 2025 review maps 28 microsampling devices, from dried spots and volumetric absorptive tips to upper-arm liquid capillary collectors, and names what a laboratory must control before their results are used: the haematocrit effect in non-volumetric dried samples, interstitial fluid from finger milking, volume, haemolysis and transport stability. Regulators on both sides of the Atlantic ask for the same two things, a device the patient can use safely and a sample fit for the test.

Targeted and untargeted metabolomics and lipidomics in dried blood microsampling: Recent applications and perspectives

This review of 2022 to 2023 work surveys dried blood microsampling for targeted and untargeted metabolomic and lipidomic profiling, finding the approach viable across paediatric research, therapeutic drug monitoring, metabolite screening, biomarker discovery, sports supervision, clinical disorder studies and forensic toxicology. The authors note that dried blood spots and VAMS dominate the published literature over other volumetric formats, and argue that harmonising analytical methods would accelerate wider clinical adoption of microsampling as an alternative to conventional plasma or serum profiling.

Blood microsampling technologies: Innovations and applications in 2022

Microsampling reduces sample volume, invasiveness, logistics and biohazard risk while improving stability and enabling at-home self-sampling, yet clinical adoption remains slow and requires standardisation and harmonisation to realise its patient-centric and decentralised potential.
