Saliva collected with the Oragene DISCOVER kit from 30 patients with pheochromocytoma or paraganglioma produced DNA concentrations comparable to paired venous blood, though with slightly lower purity from protein contamination. Saliva is a viable alternative to blood for germline screening of SDHB and SDHD mutations in this population.

    Across five extraction protocols, storage time generally reduced DNA quantity but protocol 1 (Oragene) maintained yield; DNA purity was largely unaffected by storage for commercial protocols but declined for non‑commercial methods; only protocol 1 consistently gave unfragmented DNA and protocols 1–4 yielded amplifiable DNA while protocol 5 rarely did. This supports reliable saliva self‑collection and long‑term storage for genotyping in decentralised settings.
