Evaluating the utility of storage solutions to preserve nucleic acid integrity at ambient temperature in South Africa

Moshoma et al.

The finding, in our words

The study found that DNA in blood samples stored at room temperature using commercial solutions or as dried blood spots maintained acceptable integrity for up to 90 days, supporting their use in decentralised settings where cold chain storage is not feasible. RNA integrity was below the acceptable range, and high temperatures caused degradation, highlighting limitations for ambient transport in hot climates.

Evaluation of a novel high-capacity dry plasma biosampling-lateral flow device for the testing of traumatic brain injury protein biomarkers

The DPB-LF device collected capillary blood and generated dried plasma that showed strong correlation with wet plasma for four traumatic brain injury biomarkers, with R² values between 0.83 and 0.99. Samples remained stable at 4 °C and predicted cranial lesions on computed tomography with areas under the curve of 0.70–0.76, providing a patient‑centric microsampling approach that removes cold‑chain requirements for decentralised TBI diagnostics.

A novel pretreatment of dried blood spot filter paper to improve the preservation of amino acids: a technology to increase access and care

EDTA or phosphoric acid-EDTA pretreatment of dried blood spot cards markedly improved amino acid stability under heat and humidity, maintaining accuracy for several days. This advance supports reliable self-collection and postal transport of samples for metabolic disorder monitoring in decentralised settings.

Validation of a Dried Blood Spot (DBS) Liquid Chromatography - High-Resolution Mass Spectrometry (LC-HRMS) workflow for simultaneous identification of emerging opioids

Dried blood spot sampling coupled with LC-HRMS identified and quantified 37 synthetic opioids with detection limits of 0.05 to 0.2 ng/mL. The method proved stable over seven days at ambient temperature and simplified collection, storage, and transport compared with frozen venous blood, which is pertinent for decentralised toxicology screening.

Clinical Validation of Venetoclax Volumetric Microsampling in Leukemia, with Whole-Blood-to-Plasma Conversion and Self-Microsampling Feasibility

In AML and CLL, 91% of VAMS venetoclax results fell within 20% of plasma after individualised haematocrit correction; in home sampling, 18 of 21 patients self-sampled independently and 76% of returned samples were analysable, which shows home microsampling is workable, though the authors ask for multicentre validation.
